人腦微血管內(nèi)皮細(xì)胞 HBMEC  的詳細(xì)介紹
人腦微血管內(nèi)皮細(xì)胞是血腦屏障的主要組成成分,并使其能夠限制可溶性和細(xì)胞樣物質(zhì)從血液進(jìn)入大腦。大腦微血管內(nèi)皮細(xì)胞與外周內(nèi)皮細(xì)胞相比具有一些特性:如1)腦微血管內(nèi)皮細(xì)胞有許多細(xì)胞間緊密連接,這些連接造成很高的跨內(nèi)皮阻抗,并能延遲細(xì)胞旁的通量;2)腦微血管內(nèi)皮細(xì)胞缺乏內(nèi)皮細(xì)胞窗孔結(jié)構(gòu)并且其液相物質(zhì)胞飲水平低;3)腦微血管內(nèi)皮細(xì)胞具有不對(duì)稱定位的酶和載體介導(dǎo)的轉(zhuǎn)運(yùn)系統(tǒng),從而造成“兩極分化”的表現(xiàn)型。?與外周內(nèi)皮細(xì)胞相同,大腦微血管內(nèi)皮細(xì)胞表面表達(dá)細(xì)胞粘附分子以調(diào)控白細(xì)胞進(jìn)入大腦。由于微血管內(nèi)皮細(xì)胞的器官特異性,大腦微血管內(nèi)皮細(xì)胞廣泛用于研究血腦屏障的分子和細(xì)胞機(jī)制。
ScienCell實(shí)驗(yàn)室的HBMEC分離自人大腦組織,原代凍存,冰凍運(yùn)輸。每管細(xì)胞密度超過5×105/ml。細(xì)胞特性經(jīng)vWF/Factor VIII和CD31 (P-CAM)?免疫熒光鑒定,能夠攝取DiI標(biāo)記乙?;兔芏戎鞍?DiI-Ac-LDL)。本細(xì)胞經(jīng)檢測(cè)不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell?實(shí)驗(yàn)室特制的培養(yǎng)基,可保證此細(xì)胞的良好培養(yǎng)。
